Plaque Notes | RosiePosie forms small, clear plaques. Plaque size remained consistent during the isolation process. Low titer. Optimal incubation temperature between room temperature and 30 degrees Celsius. Grew well without CaCl2, as it did with 5mM and 50mM CaCl2; however, it did not grow at 37 degrees Celsius regardless of varying CaCl2 concentrations. RosiePosie also grew well at MgSO4 concentrations of 5, 25, and 50mM concentrations when incubated at room temperature and 30 degrees Celsius. Interestingly, a clear cluster of plaques was evident for all concentrations of MgSO4 at 37 degrees Celsius for an undiluted sample of high titer lysate. In the end, plaques were clearer in the presence of magnesium. |
