Discovery NotesAfter collecting a sample from enriched soil, we successfully used techniques of serial dilution, plaque assay, full plate titers and spot titers to isolate a phage. We were able to get a spot all the way down to the 10-6 titer. This was important because we could only move on to the high titer experiments if we could get at least a 10-5 titer. A high titer experiment is important because it allows us to further analyze our phage in later experiments.
The plaques on our full plate titers are clear with a cloudy ring on the edges. All the spots are circular, with various sizes. Despite being of different sizes, all the spots have similar shape and characteristics.
Due to an accident conducting the experiment, our Gel Electrophoresis results are inconclusive.
Future plans for this project would be to get actual pictures of our bacteria. Due to the lack of TEM pictures, and inconclusive gel electrophoresis, it is difficult to affirm that we have found a new species of phage. Nonetheless, we will continue to pretend we have, and we are happy to welcome Aliel into the phage database!
