| Discovery Notes | This phage was collected from elevated bank soil that ran along Jennings Creek in Bowling Green, Kentucky. The day after the sample was collected, it was immediately enriched. In order to purify the phage, four serial dilutions were performed. The bacteriophage plaques produced were mostly turbid. In addition, the plaques were roughly 0.45cm in diameter and very round in shape. Once the one plate titer was finished, the most countable plate resulted in a titer of 1.5 x 10^2. From the one plate lysate, ten more plates were diluted out that resulted in a titer of 3.6 x 10^9. After DNA isolation, there was a DNA concentration of 70 ng/uL. Unfortunately, after this step, the gel electrophoresis did not turn out, and eventually there was not enough DNA to run a second gel. |
