Discovery NotesClyde was not originally found by me. I had to pick a plaque from my lab partner's plate and purify that because my enrichment culture did not yield any phage. My lab partner found her soil sample under a large tree on the UCSD campus in Revelle College. She dug about 1 cm deep into the soil, and the weather that day was quite warm. The temperature was probably around 80 degrees Fahrenheit.

Plaque NotesMy plaques were medium sized, clear, and haloed towards the end. In the beginning, when I first started purifying my phage, the plaques were quite big. However, as I got to the fourth purification, the plaques became smaller and the halos were very obvious. The most obvious halos were in the third purification. The plaques on my ten plate lysate were larger than I expected them to be. Overall, the plaques changed size quite often and I was not sure why. My one plate lysate plaques had a diameter of about 5mm.
