Discovery NotesSoil sampled was enrioched with Mycobacterium smegmatis + phage buffer, incubated at 37 degrees celcius for ~24 hours. 1 mL of liquid was filtered through a syringe filter, added to M. smegmatis + molten agar and incubated for 24 hours at 37 degrees celcius. An isolated plaque was picked diluted in phage buffer, added to M. smegmatis + molten agar, plated out and incubated. Purification: The plate was flooded with 10 mL phage buffer, left for hours and the liquid filtered and the lysate was prepared.
Naming NotesThe phage was isolated from garden soil in UKZN, Westville Campus. "Gardeno" comes from the phage source.
