Discovery NotesThe environmental sample was collected on September 16, 2019 from the surface of an area with light foliage. The sample was enriched and plaques were formed on a spot plate. 4 dilution series were performed in order to isolate and purify the phage from a mixed population. Small, turbid plaques along with larger, halo lysed plaques continued to be present, so a big/small test was performed to determine whether the different morphologies resulted from the same phage. It was concluded that the two plaque morphologies derived from the same phage. The big plate was flooded and a 2 more dilution series were performed in order to increase phage titer. The titer was calculated to be 2.1x10^10. A 10 plate lysate was performed to harvest a larger volume of the high titer lysate. The DNA was isolated, a restriction enzyme digest performed, and a gel electrophoresis was run. A second restriction digest was performed using different enzymes in order to get more unique banding patterns. Ahdl, BstXl, and Dralll-HF were used. Another gel was run using the second enzyme digest.
