Plaque NotesPlaques are approximately 3.3mm in diameter and turbid. Phage are more likely temperate than lyctic. (It is likely that clear plaques, in which very few or no bacteria are intact, are the result of lyctic phage. Rather, turbid plaques, where some bacteria remain intact, are likely the result of temperate phage.) Turbidity of plaques increases as temperature decreases. (42°: clear plaques, 37°: clear plaques, 30°: turbid plaques, room temperature: very turbid plaques)
Halo exists around plaques. Clear points of plaques are off center (i.e. eccentricity).
Phage does not grow in the presence of EGTA or pyrophosphate, showing that phage cannot grow in the absence of calcium or magnesium.
