Discovery NotesThe location I chose to pick my soil sample from was right outside Kindlon hall(N 41.776,W -88.096). I chose this location because The dirt was not too hard or not too moist here. It had rained the day before so finding dirt that didn’t remind me of mud was difficult so when I found the dirt it was just right in my opinion. I had filled up around half a ziploc bag of dirt, so approximately 3.5 inches worth. When we began the direct isolation, Dr. Mao had already inserted 5 mL worth of soil into the 15mL conical tube. After that, we filled the PYC broth to the 7mL line for the 15 mL conical tube as well. We left the sample to incubate for 1 hour while the sample shook vigorously at 250 rpm, ready to be used for our next lab session at 1:30 P.M.
