Discovery NotesSpringbrook Prairie Forest Preserve. The soil appeared to be very rich and fertile, surrounded by plants and trees. I dug about 8 to 12 inches into the ground. The texture was thick and smooth, but not pasty. The area had been under dry heat for the past few days.
Naming NotesI originally named my phage "Wazowski" because it was the name of one of my favorite Disney characters. After realizing it was already published in the PhagesDB.org database, I changed its name to "ShooFly" because I thought it was catchy and easy to remember as a playful expression.

Plaque NotesPhage ShooFly is lytic with a diameter of 1.0 mm. There were very few plaques yielded in the direct isolation samples. In contrast, for the following enriched isolation experiment, I yielded too many lytic phage to count. The diameter decreased from 2.5 mm to 1.0 mm during purification rounds. In which the 1.0 mm diameter occurred more and was more widespread in more of the plates. In DNA restriction analysis, ShooFly DNA concentration was 20.5 ug and needed 24.4 ul of phage DNA for ONE 0.5 ug reaction, having one visible band for the control and NspI at 10Kb.
