| Discovery Notes | This sample was collected in a moist dirt patch in Bowling Green, KY. It underwent 4 serial dilutions after being enriched. Plaque morphology was consistent in each, with the most countable plate resulting in a titer of 1.0 x 10^7 pfu/mL. Then, a ten plate lysate was completed, resulting in a high titer of 6.1 x 10^8 pfu/mL. Once the ten plate lysate was complete and calculated, the phage were then sent to an electron microscope to determine morphology. The average tail length, when measured, was 131.071 nm while the average head diameter was 47.227 nm. When the DNA was isolated, it was found to have a concentration of 60.8 ng/μL. Once the DNA was isolated, it was then subjected to gel electrophoresis and many restriction digest enzymes that revealed this phage to have a rich GC genome. |
