| Discovery Notes | A soil sample was obtained from the grounds of Albion College, and a suspension of it was centrifuged in order to separate the components by density. The liquid supernatant was extracted from the centrifuged material. We used serial dilutions of supernatant to start growing phages in smeg cultures, using both further series of dilutions and streaking to isolate a single strain of phage, which was extracted into a medium titer lysate. Once this was completed, we did a gel electrophoresis and used that sample to extract DNA and obtained an electron micrograph of individual virions. |
