Discovery Notes | I collected my soil sample from my family farm. I retrieved it from a cow pasture next to a pond. I began by isolating phages from the soil sample and enriching the sample with Mycobacterium smegmatis. I filtered the enriched soil to prepare a spot plate. I diluted the filtered enrichment to isolate only one phage from my sample. I performed serial dilutions to purify the phage. I completed 6 of these dilution series to purify my phage. After achieving a 10 fold dilution, I created a one-plate lysate to gather all of the phage particles off of a webby plate to get the maximum number of healthy phages. I also made a ten-plate lysate to increase my phage titer and ensure enough lysate to isolate the phage DNA. The titer that I used was 7.7x10^7. The results from the DNA nanodrop were 81.7ng/uL. I further analyzed Dovie by performing restriction enzyme digest with gel confirmation. |
Naming Notes | I am from Tompkinsville, KY and the one thing that my small hometown is known for is the Dovie burger. I named my phage after the restaurant Dovies. |
