Discovery Notes | My soil sample was enriched and spot plated on a bacterial lawn. Three serial dilutions were performed, but the plaques still varied in size. A big and small test was performed to ensure isolation of a single phage, and then a webby plate was plated for my one plate lysate. That lysate was then titered, collected, serial diluted, archived, and plated for my ten plate lysate. The first plating was unsuccessful, but a second performance of the plating was. The lysate was collected, filtered, serial diluted, and examined under the EM. The same lysate was also used for DNA isolation, which enabled restriction enzyme digest and gel electrophoresis to be run twice with two sets of enzymes. |
Naming Notes | I wanted to make a play on my last name (Feller) that was biology-related and rhymed. FlagellarFeller was my original idea, but exceeded the character count limit. FlagellaFella was the best satisfaction of my own stipulations for the name that met the character limit. |

Plaque Notes | My plaques were usually clustered together, with it being rare to find a single plaque who was not touching the boundary of another plaque. The plaques were about 4 mm in diameter with a roughly 1 mm in diameter clear bullseye in the center of their turbid halo. The plaques did not change in nature over time, but changed in size as the original plaques were all so squished together that it was impossible to distinguish the boundary of one from another. |
