Discovery NotesEnvironmental sample A36 underwent direct isolation and subsequently 5 rounds of purification to obtain a homogenous phage population; ultimately, a phage isolate. Soil sample A36 was collected from an area in LullWater Preserve exposed to direct sunlight, and was originally dry and sandy in consistency. After performing Direct and Enriched Isolation, A36 produced detectable plaques on the direct isolation plate that were thus picked for purification.

Plaque NotesPhage Korelly10, initially identified as A36D.1.1.1.1.1, underwent a 10-fold serial dilution in phage buffer, after which phage suspension was mixed with 100μL of host culture Arthrobacter Pascens B-1814, followed by the addition of 1mL of phage buffer (10% glycerol) and 3mL of 0.3% PYCa top agar for a final agar concentration of ~0.25%. The observed plaque morphology is on average 1mm in diameter (range 0.3mm-1mm, n=5), slightly hazy, not haloed, relatively circular, and hazy bordered.
