Discovery Notes | Isolated phage found by Dana Waxenberg in Rye, New York. The soil was collected at surface level (approximately 1-5 cm in depth) in the front yard of a suburban neighborhood. The soil was wet (sample obtained 24 hours after rainfall) and the surrounding temperature was approximately 34 degrees Fahrenheit (1 degree Celsius) and sunny. |

Plaque Notes | After 24 hours of incubation at 37 degrees Celsius, the resulting plaques were approximately 1-2 mm in diameter. These plaques were turbid with thin halos around the border of each plaque (somewhat of a bullseye look). After 48 hours of incubation, plaques exhibited similar characteristics (turbid with slightly larger halo borders), but were 3-4 mm in diameter. Titers were executed using plaques from the 72 hour incubation assay, yielding plaques of similar morphology (1-2mm in diameter after 24 hours, 3-4mm in diameter after 48 hours). After EM imaging, it was determined that the phage is a Siphoviridae with a head to tail ratio of 1:4. The head is icosahedral in shape (although it appears spherical in the EM image). Characteristics of the tail are difficult to determine at this point, due to the limiting nature of the image's quality. |
