Discovery NotesSample was collected at coordinates 42.66823 N, 71.12318 W. The sample was liquid from a puddle of water, so a 15mL conical tube was used to collect the water. Latex gloves were worn so bacteria from my skin wouldn’t contaminate the sample. The tube was dipped into the puddle and once it was full, the cap was screwed on and put into a clean sandwich bag and sealed in case of spillage. The sample was a bit cloudy, with some larger visible particles floating around. Sample was collected at 8:30 AM on 9/25/2023. The average temperature that day was 14 degrees C, and it was drizzling at
the time of collection, as well as having been raining for multiple days prior.

Plaque NotesAfter taking the Gen 1 plates out of the incubator, we observed that the 10-1 plate had too many plaques to count but they were all distinguished from each other, and they had a variety of different sizes and opacities. The 10-3 plate had 4 plaques, and all were
around 2mm in diameter and cloudy. And the 10-5 plate might have had 1 plaque, but it was very small and very cloudy so it was hard to tell. The plaque we chose on the 10-3 plate was 2mm in diameter and cloudy, so we can infer that it was in the lysogenic cycle. It didn't have a defined ring around the edges, so it was difficult to tell where the plaque ended and the rest of the agar began.
