| Discovery Notes | The soil sample was taken from moist soil below black mulch and pine needles in a hydrangea garden in a suburb of Ottawa (Barrhaven). The soil temperature could not be measured, but the ambient temperature was 13.6 degrees celsius. The soil sample itself was taken from a depth of around 5cm beneath the soil surface. Genomic DNA was sequenced on a ONT Minion, R10.4.1 chemistry using rapid barcoding V14 and PCR amplification. The longest read was 33,241 bp with 84% query cover, 88.95% identity to phage Bumble (cluster FH). Additional reads of 15,651bp had 94% query cover, 90.43% identity to phage Bumble (cluster FH). Further analysis in the lab showed that Lloydstein’s genome is almost identical to that of phage Circuit. However, the Circuit complete genome has not yet been uploaded to the NCBI database, so an exact percent identity cannot be confirmed from that site. This sequence appears to be incomplete since the longest contig is not within 10% of the completed phage circuit (cluster FH) sequence. |
