Discovery Notes | Due to Mycobacterium smegmatis being known to grow in freshwater environments and near bodies of water, the phage was collected from murky creek water. After enrichment, the phage underwent four serial dilutions. The plaque morphology appeared very consistent. After the one plate lysate was completed, the most countable plate resulted in a titer of 1.7 x 10^11 pfu/mL. A ten plate lysate was completed and resulted in a titer of 4.9 x 10^10 pfu/mL. Through electron microscopy, the tail was found to have an average length of 240.5 nm and an average capsid diameter of 68.5 nm. DNA isolation of the phage yielded a DNA concentration of 227.4 ng/uL. With the isolated DNA, restriction enzyme digests and gel electrophoresis was set up. The gel revealed that Nymph has a very GC rich genome. |
