| Discovery Notes | My phage was collected at night from a soil sample 1-inch below the surface outside of the apartment in which I reside. The soil was still slightly moist due to a recent sprinkler cycle. 1X 7H9 Complete was used for enrichment purpose. Contamination was seen in my original spot-test, but was likely a result of my own improper aseptic technique in plating as no contamination was observed in a subsequent spot-test. Once my phage was isolated, I observed that the plaques it produced were large and somewhat cloudy, signifying that I likely isolated a temperate phage. |
