Discovery NotesThis sample was taken at approximately 10:00 PM on September 25, 2016 from soil surrounding a dormitory on the campus of Western Kentucky University. The soil was shaded, surrounded by several large trees and covered by the overhang of the dormitory's roof. The temperature of soil isolation was approximately 23.9 degrees Celsius. The soil was moist, grainy, dark in color and loose, but not muddy. While many trees and shrubbery surrounded the sample, waste such as cigarette stubs, food, and paper products were present as well.

Plates were incubated at 30 degrees Celsius.

Note: Because of a low DNA concentration and a problem with gel electrophoresis,I had to repeat DNA isolation. To complete this, I also had to create new lysate and obtain a new titer number. The titer I used for EM (9x10^9 pfu/ml) was from the initial lysate. However, the titer from the new lysate (2.2x10^9 pfu/ml) will be used in the restriction digest and gel electrophoresis. The titer date listed is of the first titer. The second titer was completed on 11-15-16.

Note: I worked with a phage isolated by my classmate, Chloe Lindsey, as none of my own samples yielded plaques.
Naming NotesThe name Pemberley, Mr. Darcy's grand estate in Jane Austen's Pride and Prejudice, reflects my love of the Regency era, and the union of the humanities and science.

Plaque NotesPlaques are turbid "halos". The center of the plaque is slightly less turbid and clearly defined, while the outside of the plaque is more turbid and undefined, creating a "halo" appearance. Plaques range in size from approximately 2 mm - 2.5 mm.
