Discovery NotesThis phage was isolated from a mulch sample collected from a garden bed in the city of Pittsburgh. The sample produced plaques after several rounds of purification, with early plates showing heavy clumping and occasional contamination. Through repeated picking from isolated plaques, mostly from the 10⁻¹ dilution, a clean phage population was obtained. A webbed plate was eventually produced, allowing lysate collection and further characterization.
Naming NotesThe phage was named Persevera to reflect the persistence required throughout the isolation process, including multiple rounds of re-plating and troubleshooting to obtain a clean phage sample. The name comes from the word perseverance, symbolizing the repeated efforts needed to isolate and purify this phage. The discovery process involved overcoming contamination, clumped plaques, and multiple restarts, making “Persevera” an appropriate representation of its journey.

Plaque NotesPlaques appeared moderately sized and generally circular, with some early clumping observed in low dilutions. Later purification rounds produced clearer, more isolated plaques. On higher-dilution plates, plaques displayed consistent morphology with distinct edges, enabling accurate titer calculation.
