Discovery NotesMy phage was found on August 21, 2015. The sample was dark brown crumbly soil, collected approximately two and a half centimeters deep, in a non-populated area close to the McKee building on Western Carolina’s campus. The temperature was approximately 24.4°C and the GPS coordinates of the location is 35° 18’ 42” N 83° 10’ 49” W. One neat thing about my sample was that after it was centrifuged there was a small worm in my sample. My plaques were tested by both enrichment and direct plating. Refer to “Direct Plating/Phage Enrichment” protocol to see the process of both. I spot tested three spots from my direct plating and three spots from my enrichment. After my spots were tested one of them appeared positive for phage. My positive spot originated from my enrichment 100 plate. The plaque that appeared positive was small and clear, so it appeared to be lytic. I had discovered phage in my first soil collection.
Naming NotesI named my phage the Lytic Lemur because on the EM images that were taken, the long tail with protein rings appeared similar to those of a ring-tailed lemur.
