ProgramScience Education Alliance-Phage Hunters Advancing Genomics and Evolutionary Science
From enriched soil sample?Yes
Isolation Temperature22°C
GPS Coordinates33.80146 N, 84.3252 W Map
Discovery NotesSoil was sampled from 3" deep in the ground next to the tree adjacent to the "Additional Parking" sign at the Emory Point Conference Center Hotel North Parking entrance behind Emory Point Apartments building 6. Sandy/dry soil, no plants, covered in dead pine needles. Phage underwent 4 rounds of purification (picking and re-plating) to obtain a homogenous phage stock.
Naming NotesMy name is Fardeen Kawsar and SardineQuasar sounds kinda similar.
Sequencing Information
Sequencing Complete?No
Genome length (bp)Unknown
Character of genome endsUnknown
Fasta file available?No
Characterization
ClusterUnclustered
Subcluster--
Lysogeny NotesEfficiency of lysogeny (EOL) assay yielded 0.00025% (18 out of 7.2*10^6) which falls outside the typical lysogeny range of 0.1-100%. Therefore the phage is presumed to be lytic.
Annotating InstitutionUnknown or unassigned
Annotation StatusNot sequenced
Plaque NotesUnder the following plating conditions: 100µL of phage solution is sorbed with 100µL of host bacteria Arthrobacter pascens B-1814 and combined with 4mL of 0.4% PYCa top agar to make a full double-layer plate.
Average plaque diameter 0.8 mm (range 0.5-1.0 mm, n=10)
Plaques are clear, circular, with typically hazy edges. Sometimes a small "island" of bacteria occurs in the center of the plaque.
MorphotypeSiphoviridae
