| Discovery Notes | A volume of approximately 20 mL of moderately moist soil was collected into a sterile, 50 mL conical tube using a sterile scoop. The soil was collected from a small compost pile with an approximate area of 4ft squared located directly besides a tool shed on the east campus of Southern Connecticut State University, at 7:35 am on 9/28/2016, sunrise occured at 5:46 am. Titer of HTL equals 3.5x 10^9 DNA Concentration: 69.12 ng/ul The soil was black/dark brown and collected from an organic-rich area. The bottom layer of the compost pile contained mostly straw and a small amount of decomposed leaves scattered around. Above the layer of straw was top soil, as well as uprooted green grass(aprx. 6 inches in length,no signs of decomposing), 2 tree branches with a small girth approximately 4 ft tall,various roots, and various uprooted shrubs with varying levels of decay(little/no decay-brown/dried out). The average height of the pile was approximately 1 ft. with a few shrubs/tree branches protruding to a maximum height of approximately 4 ft. Temperature from Accuweather reported 20.56 degrees Celsius at time of collection (69 degrees Fahrenheit). Conditions featured a cloudy overcast with a slight drizzle which lasted approximately 10 minutes directly before soil was collected. Using data from accuweather, the average high and low temperatures from 9/22/2016-9/28/2016 was 21.67 degrees Celsius and 10.56 degrees Celsius respectively. Weather.com reported a total of 0.33 and 0.01 inches of precipitation on 9/27/2016 and 9/28/16 respectively, with no other precipitation recorded dating back to 9/22/2016. No wind was observed during collection, data from wunderground.com recorded a wind speed of 7 mph for 9/28/2016, and an average wind of 6.71 mph from 9/22/2016-9/28/2016. Wunderground.com listed an average humidity of 84% on 9/28/2016, with the daily minimum at 65% and maximum at 100%. The average humidity from 9/22/2016-9/28/2016 was 63.85%. |
