| Discovery Notes | The soil sample was collected near a shed where, unfortunately, a wild rabbit that had passed on had been buried several years earlier. This may have enriched the local environment and promoted diverse microbial, faunal, and phage activity. Soil can be described as dark, loamy, and moist forest soil. The ambient temperature at the time of picking was 22°C. Genomic DNA was sequenced on a ONT Minion, R10.4.1 chemistry using rapid barcoding V14 and PCR amplification. The longest read was 33,166bp with 83% query cover, 90.74% identity to phage Bumble (cluster FH). Additional reads of 14,108bp had 93% query cover, 89.80% identity to phage Bumble (cluster FH). |
| Naming Notes | Phage was named after the wild rabbit that was sadly buried in the back of my backyard by the shed. I believe that the additional organic matter provided from it may have resulted in a higher phage count. |
| Sequencing Information | |
| Sequencing Complete? | No |
| Genome length (bp) | Unknown |
| Character of genome ends | Unknown |
| Fasta file available? | No |
| Characterization | |
| Cluster | Unclustered |
| Subcluster | -- |
| Lysogeny Notes | Day 1 of lysogen testing was done. With the results the calculation based on the fact that the Lysate plate with the –2 dilution had 14 colonies counted, and the host plate at the –5 dilution had 104 colonies. The calculation: EOL= (0.014/104)*100 = 0.0135%. As per the protocol, this value is below 0.1%, therefore my phage is not lysogenic. |
