| Discovery Notes | The phage Willica was adopted from Jack Gillis, a member of the lab who originally isolated it. Jack collected his soil sample from a garden bed in his backyard, where the soil was loamy and nutrient-rich, providing favorable conditions for bacteriophage discovery. The soil sample supported the isolation of phages capable of infecting Mycobacterium smegmatis. Although I did not perform the initial sampling, I continued with isolating, the characterization and imaging process for this phage as part of my research. |

| Plaque Notes | I observed numerous small to medium-sized plaques scattered across the Mycobacterium smegmatis 7H10 plate. Most plaques were clear with well-defined edges, indicating strong lytic activity. Several plaques exhibited subtle concentric rings, resembling a bullseye pattern, which could suggest multiple rounds of infection or lysis within the bacterial lawn. There was no visible filling-in of the plaques, and I did not observe any obvious spontaneous clear plaque mutants. Based on these observations, it appears that the phage I amplified maintains consistent lytic behavior. |
