Discovery Notes | Soil sample collected from field in front of Baker Hall at Carnegie Mellon University in Pittsburgh, Pennsylvania. Soil had been fertilized earlier on 09/07/2010. Sample was collected approximately 3 inches deep into ground. Soil was moist. Temperature was approximately 80° F. The weather was sunny and there had been no recent rainfall. |

Plaque Notes | Plaques are approximately 3.3mm in diameter and turbid. Phage are more likely temperate than lyctic. (It is likely that clear plaques, in which very few or no bacteria are intact, are the result of lyctic phage. Rather, turbid plaques, where some bacteria remain intact, are likely the result of temperate phage.) Turbidity of plaques increases as temperature decreases. (42°: clear plaques, 37°: clear plaques, 30°: turbid plaques, room temperature: very turbid plaques) Halo exists around plaques. Clear points of plaques are off center (i.e. eccentricity). Phage does not grow in the presence of EGTA or pyrophosphate, showing that phage cannot grow in the absence of calcium or magnesium. |
