Discovery NotesIsolated by TA Dominic Garza.

Mixed culture used for isolation:
An enriched culture was prepared by combining 15 mL of soil with 25 mL of PYCA broth. The bacterial strains of A. sp., A. globiformis, A. sulfureus and A. atrocyaneus were then added in a ratio of 1:5:192:588 respectively. The enriched culture was incubated at 30℃ for 48 hours with shaking. After filtering with a 0.22 µm filter, the sample was plated on PYCA plates with 2X PYCa soft agar using the standard pour plate method.

Sample description: The soil sample was moist. The collection site was rural and far away from any buildings; however, there was quite a lot of foot traffic considering it was taken from along the Eaton Canyon hiking trail. There was a waterfall and stream close to the soil collection site.
