| Discovery Notes | The soil sample used to isolate this phage was taken from a dry area next to a light post, approximately 1.5 cm deep in soil. The outside temperature was 20 degrees Celsius, and it was sunny outside. This phage was isolated from a lysate made from an enrichment that contained both Mycobacterium smegmatis and Arthrobacter. I had hoped to isolate a unique phage by adding the selective pressure of two hosts. Ideally, I wanted to isolate a phage that infects both hosts, but Nick only infects M. smeg. Because the ideal temperature for growing Arthrobacter bacteria is 30 degrees Celsius, I incubated the enrichment at 30 degrees Celsius, and used that temperature to incubate M. smeg. as well during isolation |

| Plaque Notes | Plaques formed by Nick are 2-3 mm in diameter and turbid. Plaques form after 24 hour incubation at 30 degrees Celsius, but become more defined after a 48 hour incubation at that temperature. Plaques also form when the incubation temperature is 37 degrees Celsius or room temperature. |
