Discovery Notes | Near a pond located on a family-owned farm where cattle are raised. The collection site is surrounded by a large pond therefore the soil is always relatively moist. The farm in which the soil was collected is occupied by approximately 80 head of cattle. The site is located far from any roads or buildings. Surrounding the site is a tree line with brush, weeds, green grass, dead leaves, limbs, and cattle feces. At the time of collection, the soil was moist due to being located near a body of water. |

Lysogeny Notes | When incubated at 30 and 37 degrees Celsius my phage formed a lysogen. Mesa could be observed where 10^0 and 10^-1 dilutions were spotted on a spot plate. Mesa is a lysogenic Mycobacterium smegmatis that did not get lysed by the bacteriophage. Mesa can be seen on the plates as a turbid plaque surrounded by a zone of clearing. This indicates that M.smeg has some resistance to the phage. With further testing, my lysogen showed results indicating that I have an unstable lysogen. It appears my phage has lyric and lysogenic properties. |
