| Discovery Notes | I collected soil from West Arlington, Texas at 4:30pm. The temperature at that time in the afternoon was 96F with 43% humidity. The location was in a flowerbed to the right of my yard facing the house, next to a tree. I dug approximately 1.5 inches below the surface, collected the sample, and placed it into a plastic bag. |

| Lysogeny Notes | I obtained 5 plates and labeled them with the name of the phage, the name of the host bacterium, and one of the following five dilutions: 10^2, 10^3, 10^4, 10^5, 10^6. I then added 100μl of my high titer lysate phage sample to each of the plates and spread them evenly. The TA then provided me with the dilution series of host bacteria. I then added 100μl of host bacteria to the corresponding dilution (100μl of the 10^2 dilution to the plate labeled 10^2) and spread the solution evenly on top. The plates were then incubated for the duration it takes colonies to form (typically 2-5 days) Results were negative |
