Discovery Notes | Weather Conditions at the time of collection (about 4:17pm): Mostly cloudy with 7 mph wind, temperature was around 62oF, humidity 57%. During the day: Partly cloudy with a high of 38o F and a low of 32o F, light winds and sunny at times Surrounding vegetation included straw pieces, grass, twigs, and rocks. Soil was transported approximately 30 minutes at room temperature before storing in a refrigerator for 18 hours, then was transported for approximately 3 hours in my backpack at room temperature; moist consistency contained some grass and straw pieces |

Lysogeny Notes | While lysogen testing has not been performed yet, the morphology of the plaque suggests the phage is lysogenic. Also, no plaques were found initially in the spot tittering. I hypothesized that this may be due to the great amount of lysogen being produced by the phage. In order to test this, I added 150 uL less of host cell to the spot titer. By using less host cells, the stationary phase will be prolonged and the phages will be able to infect the host cells for a longer time and not produce as many lysogens. This proved as a success and plaques were found. It also is further cause to believe the phage exhibits lysogenic characteristics. |
