| Plaque Notes | Our plaques began in two different sizes and morphologies, but became more uniform after our last purification and plate webbing. Starting, there were very small clearings, around 1 mm in diameter, almost appearing like dots. There were also larger plaques, around 4 mm in diameter, with halos and a more turbid appearance. During our third round of purification, it was hard to tell the plaque morphologies because we had an issue with the diluion of our plates. The concentration of 10^0 was the only one with plaques; however, it was plaqued out, so one could not obtain a morphology. The other plates did not have plaques on them so there must have been an error made while making the dilutions. However, we were able to use the plaqued out plate to make our lysate. From the lysate, we were able to make eight high-quality diluted plates. On these plates, we could see the plaques better and were able to observe their morphologies. The plaques were uniform this time, all around 2 mm in diameter and clear wih no halos. |
