Naming Notes | IchabodCrane is a Siphoviridae with a distinguishable long and flexible tail. Capturing intact phage for imaging was complicated because its lengthy tail and capsid were severed throughout the sample. The slaughtered scenes were reminiscent of The Sleepy Hollow tale of lanky and headless Ichabod Crane. The time and wooded location in which the phage was discovered also befitted the Halloween season. |

Lysogeny Notes | Lysogen tests were observed over 10 days. Mesa were not found. The tests were repeated twice to ensure that the lack of mesa was not due to other factors. Mesa were still not found which strongly suggests that IchabodCrane is a virulent bacteriophage for S.scabiei. |

- Clear, distinct, symmetric circles
- Plaque clusters will form due to phage traversing the agar and infecting nearby cells. It appears as if some plaques are larger than individual ones but they are usually two plaques infecting bacterium close in proximity.
- Plaque assay (at 20 hrs): 1.1 mm in diameter with clear edges under 28.4 degrees centigrade incubation
Note: When plaque assays have extended (28 C) incubation of 52 to 120 hours, or transferred to 4 C storage within 48 hours of incubation, the plaques will grow 1.9 mm in diameter with faint to cloudy edges. |
