Discovery NotesThe soil sample from which phage Ruriko was isolated was collected at 7:25 am CST on August 26, 2025, from a compost bin in the backyard of a home in rural Lindale, Texas. The morning was partly cloudy with an ambient temperature of 22 °C. Clearing away a layer of decaying fruit and leaves revealed moist soil teeming with worms, bugs, larvae, and the remnants of banana peels, watermelon rinds, corn husks, and coffee grounds. A plastic spoon was used to make a 4-inch wide and 3-inch deep hole, scoop a soil sample into a Ziploc sandwich bag, and take it to the laboratory for processing that morning. Five grams of the soil sample were weighed out and used for further processing, using the enriched isolation method.
