Discovery Notes | Phage amplification was performed by making an enriched culture. 25 mL of PYCa and 4 mL of 48 hour mixed culture of Arthrobacter was added to the soil sample, which was subsequently incubated at 30ºC for 48 hours. Following filtration by using a 0.22 µm syringe, a 1-plate plaque assay was performed on Arthrobacter species, A. atrocyaneus, A. sulfureus, and A. globiformis to determine if enriched isolate contained phage. Plaques were found to be most prominent on A. sulfureus, which was used as the host bacteria for all subsequent experiments.
