Discovery Notes | The water sample was collected from a shallow part of a creek (4-5 inches deep) which streamed off from the Barren river. The temperature when collected was 34 degrees C. The humidity was 62% and the surrounding soil was moist as it had recently rained. The water sample was enriched and spot tests were used to determine if phage was present. An initial serial dilution was performed and plated. Purification of the water sample was done by picking a single plaque from the 10^-4 plate of the initial plated serial dilution and plating another serial dilution from the picked plaque. From the second serial dilution, a single plaque from the 10^-4 plate was picked, serially diluted, and plated. This process was repeated one more time. The webby plate from this final plated serial dilution was flooded with phage buffer and filtered to produce a high-titer lysate. |
Naming Notes | I had just learned what the word cornobble meant, to slap someone with a fish, and I thought it was funny and fitting because the phage came from a river water sample. |
